Chromosome rearrangements involving the MALT1 (MALT lymphoma-associated translocation 1) gene on chromosome 18q21 have been observed in several types of lymphoma. In t(11;18) (q21;q21), a well-documented translocation involving the MALT1 gene, a gene fusion is produced on the der(11) chromosome between a 5' portion of the API2 (11q21) gene and a 3' portion of the MALT1 gene (18q21). The resulting API2/MALT1 fusion gene encodes an abnormal, but functional chimeric protein. Hybridization with the LSI MALT1 (18q21) Break Apart Rearrangement Probe will identify t(18q21) but not the specific translocation partner. The probe will also identify aneuploidy of chromosome 18.

The LSI MALT1 Dual Color, Break Apart Rearrangement Probe consists of a mixture two FISH DNA probes. The first probe, a 460 kb probe labeled in SpectrumOrange, flanks the 5' side of the MALT1 gene. The second probe, a 660 kb probe labeled in SpectrumGreen, flanks the 3' side of the MALT1 gene. There is a 65 kb gap between the two probes. The known break points within the MALT1 gene region are found in introns 2, 3, and 8.

Results of Hybridization

In a normal cell that lacks a t(18q21) in the MALT1 gene region, a two fusion signal pattern will be observed reflecting the two intact copies of MALT1. In an abnormal cell with a t(18q21), a one fusion, one green, one orange signal pattern will be observed.
